Predição in silico e validação in vitro de peptídeos antagonistas do receptor de IL-1

O processo inflamatório é uma reação fisiológica do nosso organismo, entretanto pode ocorrer a desregulação desse processo e isso está relacionado ao desencadeamento e agravamento de diversas doenças. O principal fator, que torna o processo inflamatório patológico, é a liberação exacerbada de citocinas pró- inflamatórias. Dentre elas, está a IL- 1β, que quando expressas em quantidades aumentadas, perpetuam o processo inflamatório e não permitem que o mesmo siga para a fase de reparo. A IL-1Ra é uma molécula antagonista do receptor de IL-1R1, que controla os níveis de IL-1β ao se ligar ao receptor e impedir a ligação da IL-1β. Por essas características, se tornou um alvo
terapêutico para o controle de patologias desencadeadas por IL-1. Nesse sentido, existem medicamentos, que tem como objetivo neutralizar a função biológica da IL-1β. No entanto, os inibidores de IL-1β encontrados atualmente no mercado, são de proteínas recombinantes, que possuem custo elevado, pouca estabilidade e efeitos adversos. Visto
isso, o objetivo desse trabalho foi predizer dois peptídeos, baseados na molécula IL-1Ra, com capacidade de atenuar as respostas desencadeadas pela IL-1β, através do bloqueio do receptor IL-1R1. Essa alternativa representa uma opção promissora para o tratamento de doenças inflamatórias, visto que os peptídeos são moléculas estáveis, de fácil produção e baixo custo. Foram desenvolvidos dois peptídeos: PAD12 e PAD3, através do Swiss PDB Viewer, uma ferramenta de bioinformática que possibilita a visualização da estrutura proteica de forma detalhada, sendo possível selecionar os aminoácidos que fazem interação com cada domínio do receptor IL-1R1. A seleção desses aminoácidos foi baseada na estrutura tridimensional da ligação da IL-1Ra com o receptor IL-1R1, encontrada no Protein Data Bank (PDB) com código I1RA. O peptídeo PAD12 interage com os domínios 1 e 2 do receptor, enquanto o peptídeo PAD3 interagem com o domínio 3. Após a predição, foi realizada a síntese química dos peptídeos pela técnica de Fmoc-Sinteses. A validação dos peptídeos PAD12 e PAD3, foi realizada in vitro em cultura de células imortalizadas (RAW
264.7). As células foram expostas a LPS (5ug/mL) por 24 horas e posteriormente tratadas com os peptídeos (5ug/mL) por 24 horas. O teste de ELISA, foi realizado com o sobrenadante celular para dosagem de IL-1β (pg/mL). O teste mostrou que os peptídeos foram capazes de ocasionar uma diminuição de 50% nos níveis de IL-1β. Por fim, foi realizado o ensaio de MTT, para comprovar que os peptídeos não são tóxicos para a linhagem celular.
The inflammatory process is a physiological reaction of the organism. However, the deregulation of this process can occur and this is related to the triggering and worsening of several diseases. The main factor that makes the inflammatory process pathological is the exacerbated release of pro-inflammatory cytokines. Among them is IL-1β, which when overexpressed can perpetuate the inflammatory process and impair the repair phase. IL- 1RA is an IL-1R1 receptor antagonist molecule, that controls IL-1β levels. Due to these characteristics, it has become a therapeutic target for the control of pathologies triggered by IL-1. In this sense, there are drugs that aim to neutralize the biological function of IL-1β. However, the IL-1β inhibitors currently on the market are recombinant proteins, which have a high cost, low stability, and adverse effects. Given this, the objective of this work is to predict two peptides, based on the IL-1RA molecule, with the ability to attenuate the responses triggered by IL-1β, by blocking the IL-1R1 receptor. This alternative represents a promising option for the treatment of inflammatory diseases since peptides are stable molecules, easy to produce, and have low cost. Two peptides were developed: PAD12 and PAD3, through the Swiss PDB Viewer, which allows the visualization of the protein structure in a detailed way, being possible to select the amino acids, which interact with each domain of the IL-1R1 receptor. The selection of these amino acids was based on the threedimensional structure of the binding of IL-1Ra with the IL-1R1 receptor, found in the Protein Data Bank (PDB) with the I1RA code. The PAD12 peptide interacts with domains 1 and 2 of the receptor, while the PAD3 peptide interacts with domain 3. After the prediction, the chemical synthesis of the peptides was performed using the Fmoc technique. The validation of PAD12 and PAD3 peptides was performed in vitro in immortalized cell culture (RAW 264.7). Cells were exposed to LPS (5 ug/mL) for 24 hours and treated with the peptides (5 ug/mL) for 24 hours. The ELISA test was performed with the cell supernatant to measure IL-1β. The test showed that the peptides were able to cause a 50 % decrease in IL-1β levels. Finally, the MTT assay was performed to prove that the peptides are not toxic to the cell line.
